Hello, my Arduino controller board only has 4 wires, 5v/gnd/rx/tx

how do I reset the board? GRN usually connected to the negative ???I don’t understand what you are asking. What are you trying to do with the Defusable Clock? Program it using an Arduino? If you want to program it, you need a USB to serial adapter or cable. See http://nootropicdesign.com/defusableclock/hack.html
